Questions about acetylcholinesterase inhibitors and alternatives

I have a few questions regarding acetylcholinesterase inhibitors and other nootropics that modulate choline in the brain. My goal is to enhance choline levels so that racetams will be more effective and choline levels in the brain will not be depleted as quickly. For reference, I take vyvanse 20mg 5 days a week, am prescribed pregabalin (200mg/day), and sometimes if I need to study I will add oxiracetam, pramiracetam, and/or noopept, sometimes adding a bit of matcha or Kratom as well. I take CDP choline with these to avoid excitotoxicity as much as possible.

I was looking for a list of acetylcholinesterase inhibitors and I couldn’t find it. I remember someone posted a list of herbal maois at some point, I’m basically looking for that for AChEIs. I’m looking for supplements/nootropics I can take, not medications. I’m wanting these to enhance racetams/vyvanse, which I take often. I’m also not really sure if I entirely understand the difference between acetylcholinesterase inhibitors and nootropics that modulate the acetylcholine system. I sort of understand how they work in the brain based on my biology courses, but I don’t entirely understand how they affect nootropics.

I already have huperzine-a, which I know works for what I’m asking here, but I’m just looking for other alternatives. I know noopept and emoxypine modulate choline in some way but I don’t entirely understand how, and iirc quercetin and memantine has some effect on choline, but again I don’t fully understand how that works.

My current (not complete) stack in addition to nootropics/vyvanse is: pycnogenol, co q-10, magnesium, vitamin d, b complex, cdp choline, and psilocybin mushroom microdose .1G 4-5x a week with lions mane as well.

I know this is a lot of questions but I would really appreciate any response 🙏

Supplements to take with Vyvanse?

I recently started on 30 mg of Vyvanse, and while I know that taking supplements isn’t absolutely necessary with this medication, I’d like to optimize my routine to maximize its effects and support my overall health and ADHD symptoms.

Based on my research so far, here are the supplements I’m considering:

• L-Tyrosine

• ALCAR (Acetyl-L-Carnitine)

• DLPA (D,L-Phenylalanine)

• Choline

• Omega-3s

• Vitamin D + K

• Multivitamin

• Magnesium Glycinate

I exercise daily and follow a whole-food diet with an emphasis on protein. I also drink electrolytes mixed in with plenty of water throughout the day.

Any suggestions on other supplements to add, or adjustments to make? Thanks in advance!

Why does norepinephrine upgrade my brain?

I suffer from decreased attention span after a head injury, chronic fatigue, brain fog, and ADHD, but when I take medication to increase noradrenaline, everything improves at once.

However, when I take serotonin, I feel like noradrenaline is suppressed. (For example, taking Cymbalta, an SNRI, is effective for the first month, but gradually becomes counterproductive, and when I switch to a medication that increases noradrenaline alone, I feel better.)

On the other hand, there is something that bothers me, and that is that increasing noradrenaline puts a lot of strain on my heart. My blood pressure exceeds 160 and my QT is greatly extended.

What I am wondering here is:

①Is there a way to increase noradrenaline only in the brain (prefrontal cortex?)? (Also, is there a way to reduce the side effects on the body?)

②I am looking for the root cause of chronic fatigue, and what are the possible causes involving noradrenaline? (Even though I have been diagnosed with ADHD, when I take medications that increase dopamine, they all have the opposite effect. It seems that the mechanism by which noradrenaline is produced from dopamine is not working well for me.)

  1. Is it possible that increasing noradrenaline is not the true solution, and that other brain substances or hormones that are linked to the increase in noradrenaline are important?

I am very interested in "safely" increasing noradrenaline, and I am worried that there is no other way to save myself (but increasing noradrenaline has very large physical side effects, which is troubling me. Also, for some reason, Atomoxetine is the only thing that is supposed to increase noradrenaline but does not work at all. I get side effects, but my executive function does not improve with Atomoxetine)

Bromantane Stack Advice for extreme Social Anxiety

Background:

Hi so I'm very new to all of this so I apologize if my knowledge isn't completely accurate. I'm currently suffering with pretty severe social anxiety and lack of sharpness and focus due to my ADHD. I've been trying to talk to more people in hopes of improving my social skills and lowering my anxiety but lately it's just been making me lose faith in myself and getting me more anxious. I remember hearing about Bromantane a while back and reading about on the various subs seems like it could work in terms of helping me calm down due to being Anxiolytic with some long term's effects. The only other drugs I've tried was Fluoxetine and Nicotine. Fluoxetine worked kind of for a bit it gave me a lot of fatigue and later actually made me more anxious. Nicotine actually did help however I only took it once in a form of Zyn a friend gave me. I might be overexaggerating the effects but for the first time in really long time I felt normal and calm. I was able to communicate without having to overanalyze every little thing. I of course didn't take it again because it's nicotine I don't want to develop an addiction.

Current Plans:

I'm currently taking 10mg of Ritalin and 2000mg Magnesium L Threonate. I've tried looking if there were any effects if you take Bromantane with Ritalin however I couldn't find any information. My Bromantane is coming next week it's in Solution form. (Bromantane – Solution, 1500mg). After a separate a reddit post I've decided to go with the Intranasal route with 25mg each day for 30 days. If I can't handle intranasal, I'll go with 50mg orally each day. I have been suggested make a Nasal Spray or Sublingally(The thing where you put it under your tongue awhile don't remember the spelling) by mixing it with a fat such as olive oil or MCT oil. But I'm not sure if I trust myself to be able to do it properly so I'm just going with either intranasal or orally for now.

Questions:

I need to know if I can take this medication while on Ritalin.

I also need to know if I should be taking any other Nootropics to see an actual effect. Of course, I'm not looking to rely on this soley but I really need these feeling of anxiety and stress to be dumbed down a bit if I can make any process in my social skills so if you have any suggestions on what I should get for a better effect please let me know.

Edit: I don't know if this matters but I also suffer from sentence formation and word retrieval and verbal fluency a lot. If there is something that can help with that please let me know.

Agmatine and DHEA: Unexpected Synergy or Overstimulation? Looking for Insights

Hey everyone, I wanted to share some recent observations and see if anyone here has had similar experiences with combining Agmatine and DHEA.

Background: Quick story on my context – a few years ago, I went through a pretty rough burnout that hit my adrenals hard. Since then, I've been on a recovery path, trying to balance things out, physically and mentally. Part of that journey included supplementing with 100 mg of DHEA daily, but my levels have barely moved over the years. Even on 100 mg, my DHEA-S level was hovering around 280, not really responding. But then... I decided to give Agmatine a go.

My Agmatine Experience (so far): I started with 200 mg of Agmatine and noticed something strange – it felt like the effects took a full 24 to 48 hours to kick in. It’s been one of the strongest supplements I’ve tried: big boosts in energy, motivation, emotional stability, and drive. I actually feel way more balanced with it and can even sense Agmatine’s known tolerance-resetting effects, like on caffeine or THC (which I use occasionally). Right now, I’ve scaled it down to 30 mg a day to manage the overstimulation, but the positive effects are still there.

The DHEA Change (and Side Effects): Since starting Agmatine, for the first time in 2 years, my DHEA-S has actually moved up, now sitting around 380. I can really feel the difference – deep voice, strong libido, vivid dreams, high energy, and more… plus some side effects. I’m dealing with extra oily skin and more breakouts, which I didn’t have on 100 mg of DHEA alone. I also get occasional diarrhea and am definitely feeling a bit overstimulated from time to time.

I’m wondering if Agmatine somehow amplifies DHEA’s effects or helps with its absorption. It’s known for resetting tolerances to things like caffeine and THC, so could it be “resetting” my DHEA tolerance too? It’s making me curious if there’s some biochemical interaction here that’s finally getting DHEA to work more effectively in my body.

Questions: Has anyone here experimented with Agmatine and DHEA together? Have you noticed similar effects or have any insights on why Agmatine might make DHEA so much more noticeable? I’d love to hear any theories, personal experiences, or science-y explanations. I’m kind of just baffled (but excited) about what’s happening here and would really appreciate any advice or stories from anyone who’s had something similar happen.

Thanks a ton for reading, and looking forward to any insights you can share!

9-Me-BC and 4'DMA-7'8DHF stack

Has anybody tried this stack? Most of it seems focused on 9-me-bc and semax or noopept or NSI-189. Nobody seems to have tried it with 7'8DHF or 4'DMA-7'8DHF, which are potent TrkB agonists. If I wanted to create a super nuerogenesis stack, wouldn't is make sense to add these together?

Magnesium L-Threonate Recommendation

I've been taking Glycinate, but would like to try Threonate. Which brand do you recommend? I am on Amazon Canada looking at the following three brands: NOW, Naka and New Roots. For all three, each capsule contains about 50mg of elemental calcium.

A second question is what happens if I am also taking GABA supplements?

Question about Modafanil, Adderall, Vyvanse Need Advice!

I’ve struggled with CFS/ME (and probably undiagnosed ADD) for over a decade. At the point where I cannot get through the day without stimulants. (Yes, I am well-versed in pacing and PEM and all the potential problems with stimulant crash, but until a better treatment is found I need these medications to function and care for my family even though a “good day” for me is probably 25% of a normal day for most other people.)

So my question is about stacking. I was doing reasonably well on Vyvanse, but it stops working as well and I did not want to increase my dose past 40 mg. Also, I was noticing that it was giving me waves of - not sure how else to describe but , dysphoria? Does that make sense?

In any case, I wanted to try some thing else and my doctor recommended modafinil. I’ve been on a low dose (50mg) and am supposed to increase to 100. The problem is the modafinil is giving me nothing as far as energy. I am wondering if it would be more beneficial to stack it with either Adderall or Vyvanse?

Please share any stacking, supplement and or cycling (alternating meds on different days) that has worked for you. Thank you.

Bromantane and 9-me-bc

Can i stack these? Have some leftover bromantane and i’m starting 9-me-bc. I also have a new pack of methylene blue. I suppose that’s fine to add? Using L-tyrosine and ALCAR as support for bromantane/9-me-bc. Anything to add/remove or other tips? I have alot of stuff at my disposal, and can get basically everything home.

Magnesium and Vitamin D at night causes insomnia?

I started taking magnesium and vitamin d at night before bed. Usually, I only take it in the morning, but I read these two can be taken together at night for improved sleep. I found the opposite to be true. Does anyone know why? I take half the magnesium capsule in the morning, and half at night. So that's 100mg in the morning, and 100mg at night. It doesn't seem that much. Why does it have this insomnia effect. I spend time rolling around in bed instead of falling asleep. I always take it with fat, like cheese or flaxseed oil. Is it the Vitamin D or Magnesium that's keeping me up? What if I keep the Vitamin D for mornings only and take Magnesium at night? Or should I not take any vitamins or minerals at night at all?

Struggling to understand Magnesium Glycinate

I've been reading conflicting things about Magnesium Glycinate, some say you should take 600mg/day, others say max daily dose for men is 420mg/day so I started taking Magic Magnesium because the front made it look like 600mg of Magnesium Glycinate but now reading the back there's only 240mg of Magnesium Glycinate per 2 tablets which is how much it tells you to take so I was looking to change brands but I'm struggling to understand what a good brand and dose is because I'm looking on Amazon and there's products like these that claim to have 1000mg of Magnesium Glycinate in them which jusr doesn't make sense with the doses I'm reading and makes me not trust the products. Can anyone help me with understanding what dose I should be taking and what brands to look out for cause I'm just lost with it all. My goal for taking Magnesium Glycinate is mainly to help with nerve damage and a slipped disc. These are the other products I'm taking as well so if anyone has any other comments or suggestions on any of them or other supplements that would help I'd be happy to hear them, really just want this pain to go away ASAP

am i doing more harm than good?

I’m a 30yo F, dx include adhd, major depression, experiencing serious burnout from being a single mom. i usually lose a lot of energy midafternoon and my motivation and concentration levels are so low that i am just able to do the bare minimum in keeping my household run smoothly. because of my impulsivity, i tend to jump on supplements thinking if drugs.com doesn’t explicitly mention any adverse reactions, then they’re safe.

first thing in am: 120mg duloxetine 60mg lisdexamphetamine 5000 iu vitamin D women’s probiotic 2500 mg collagen peptides

after eating (usually 3-4 hours later): 1200 NAC

3-4 hours after that: women’s multivitamin (to replete any minerals from the NAC)

before bed: 50mg trazodone

~1mg phenibut 3-4x a week when i have a busier day.

i’m considering adding NMN to the stack but i’d like some advice about what i’ve got going on now.

TIA.

Which stack can I safely add when using Pregabalin & Kratom?

I use to take my prescribed Pregabalin 100-300mg if needed and Kratom almost daily in the last 2 weeks (sadly, I definitely am reducing atm, planning to quit).

I am very interested in supplements and herbs and have many at home to try but am worried about possible risks or dangerous interactions with the Pregabalin or Kratom or both together.

Maybe someone here more knowledgeable about pharmacology or biochemistry can give some advice on what would be okay and would be rather potentially risky. It would be nice to add some stack that is possible to take with my kratom/pregabalin in the beginning (without risky interactions) so I am able to take less and less kratom/pregabalin for my depression and anxiety disorder.

Following supplements/herbs I have laying around but never got to try:

  • Gotu Kola

  • Bacopa

  • Lemon Balm

  • Melissa (extrac) - i guess Lemon Balm

  • Passionflower

  • Blue Lotus

  • Wild Lettuce (10:1 extract)

  • Magnolia Bark

  • Mulungu (plain herb)

  • Silexan (Lavender standardized extract)

  • Rhodiola Rosea (extract „WS“?)

  • Taurine (1-3g and what about higher doses like up to 6-8g?)

  • L-Tyrosine

  • Mucuna Pruriens

  • Muira Puama (10:1 extract)

  • Ashwagandha

  • Black Seed Oil

  • Hydroxyzine (antihistamine medication)

  • SAM-e

  • Gingko

  • Ginseng

  • GABA

Anything to remove fear for a 85yr old?

Anything to remove fear for a 85yr old?

My friends grandma has a lot of fear that’s what they told me

‘She is extremely afraid of being alone, even in the presence of the family. She feels extremely anxious and trapped, especially in an unfamiliar environment. She feels like a prisoner. There is a strong pressure on the head and chest. Became very forgetful. Please support quickly, thank you!’

So I suggested propranolol but maybe there’s something else? Also what would be the recommended dosage?

Docs here are mostly useless and just prescribed her an anti depressant.

She also needs a mood enhancer along with something for fear but age is also a factor so I don’t want to recommend something which I might recommend to someone younger as she might not be able to tolerate it or get side effects

Let me know

Thanks

What can I add to my stack for mood?

Which additional supplements with different mechanisms of action could I add to improve mood? I'm 22 yo male. I already take the following supplements, with their respective mechanism of action.

Multivitamins and minerals:

  • Provide essential nutrients that act as cofactors for enzymes involved in neurotransmitter synthesis
  • Support overall brain function and cellular energy production

Acetylcysteine (NAC):

  • Increases glutathione levels, reducing oxidative stress in the brain
  • Modulates glutamate signaling, potentially reducing OCD symptoms
  • Affects dopamine release and uptake, which may help with addiction-related behaviors

Bacopa monnieri:

  • Enhances neurotransmitter release, particularly serotonin and dopamine
  • Increases dendritic branching, potentially improving neural plasticity
  • Acts as an adaptogen, helping the body resist stress

Iron:

  • Essential for dopamine synthesis and metabolism
  • Supports oxygen transport to the brain, enhancing cognitive function

Zinc:

  • Modulates NMDA receptor activity, potentially reducing anxiety and depression
  • Acts as a cofactor for enzymes involved in neurotransmitter synthesis

Electrolytes:

  • Maintain proper neural signaling and brain function
  • Support hydration, which is crucial for optimal cognitive performance

L-theanine:

  • Increases alpha brain wave activity, promoting relaxation without sedation
  • Modulates GABA, dopamine, and serotonin levels in the brain
  • May enhance BDNF (Brain-Derived Neurotrophic Factor) expression

Choline Bitartrate:

  • Precursor to acetylcholine, a neurotransmitter involved in memory and mood regulation
  • Supports the structural integrity of cell membranes in the brain

Vinegar:

  • May help stabilize blood sugar levels, potentially reducing mood swings
  • Contains acetic acid, which might influence gut microbiota, indirectly affecting mood through the gut-brain axis

Healthy eating (antioxidants, macronutrients):

  • Provides essential nutrients for brain function and neurotransmitter synthesis
  • Antioxidants combat oxidative stress in the brain
  • Balanced macronutrients support stable blood sugar and energy levels

Coffee:

  • Blocks adenosine receptors, increasing alertness and potentially improving mood
  • Stimulates the release of dopamine and norepinephrine

Fish and fish oil:

  • Provide omega-3 fatty acids (EPA and DHA) that support cell membrane fluidity in the brain
  • Modulate inflammation and oxidative stress in the brain
  • Influence neurotransmitter function and neuroplasticity

Olive oil:

  • Contains polyphenols that may reduce neuroinflammation
  • Supports overall brain health through its antioxidant properties

Coffee fruit extract:

  • May increase BDNF levels, potentially supporting neuroplasticity and cognitive function

Rhodiola rosea:

  • Acts as an adaptogen, modulating the stress response system
  • May influence serotonin, norepinephrine, and dopamine levels in the brain

Phosphatidylserine:

  • Supports cell membrane function in neurons
  • May modulate cortisol levels, potentially reducing stress response

Venlafaxine:

  • Inhibits the reuptake of serotonin and norepinephrine, increasing their availability in the synaptic cleft
  • At higher doses, also affects dopamine reuptake

Mirtazapine:

  • Blocks α2-adrenergic autoreceptors, increasing norepinephrine and serotonin release
  • Antagonizes certain serotonin receptors, potentially reducing anxiety and improving sleep

Ketamine:

  • Blocks NMDA receptors, potentially leading to increased glutamate signaling
  • May rapidly increase BDNF levels and promote synaptogenesis

Hatha yoga:

  • Activates the parasympathetic nervous system, reducing stress response
  • May increase GABA levels in the brain, promoting relaxation

Pranayama:

  • Influences the autonomic nervous system, potentially reducing stress and anxiety
  • May affect neurotransmitter levels through changes in brain oxygenation

Meditation:

  • Alters activity in the default mode network, potentially reducing rumination
  • May increase gray matter density in brain regions associated with emotional regulation

Exercise:

  • Increases BDNF levels, promoting neuroplasticity
  • Stimulates the release of endorphins and endocannabinoids
  • Modulates the HPA axis, potentially improving stress response

8h of Sleep:

  • Allows for synaptic homeostasis and memory consolidation
  • Supports glymphatic system function, clearing metabolic waste from the brain

Light box therapy:

  • Regulates circadian rhythms by influencing melatonin production
  • May affect serotonin levels and activity in mood-regulating brain regions

I've had negative experience with creatine, ashwagandha and high dose psilocybin.

Agmatine and benzos, advice from an expert 🙏

So i am in a deep dark benzo hole, dual addicted to diaz and alp

i know i have to stabilize on diaz and taper off that but would it be a bad idea to take agmatine during the process? or would it act as a potentiator and make things worse? i wasnt able to find much info online about this and apparently its banned some places, i dont know for what reasons.

anything else that could possibly be of aid? im thinking lions mane would probably help prevent BIND?

i know i should be in a hospital setting, im trying to get in asap.

Dlpa, tyrosine, Alcar, and B complex

Currently taking DLPA 500mg, L-Tyrosine-500mg, ALCAR - 500mg, and B-Complex for a week.

I've found I have more energy throughout the day. Not as stimulating as coffee (which is okay) and also doesn't have the drop down I sometimes get with coffee. It's consistently throughout the day. Definitely elevated mood (probably from the DLPA?).

Today, I didn't take them in the morning to see how it feels. Definitely feel groggier but I also didn't workout this morning so that could attribute to it.

Questions I have are:

  1. What's the frequency of taking these 4 together I can do safely without withdrawal symptoms, as well as tolerance?
  2. Anything I should be experimenting to remove from the stack, and/or replacing (I also have NALT).
  3. Anything I should be adding to offset/supplement/increase absorption of the stack?
